Ionizing radiation (IR) therapies stimulate both innate and adaptive immune cells present in the tumor microenvironment and elicit an antitumor response at distance of the irradiated tumor sites. This biological response by IR is called the ‘abscopal’ effect. IR induces immunogenic cell death in tumor cells, leading to the release of tumor antigens and damage-associated molecular patterns(DAMPs), which in turn prime and activate antigen-presenting cells (APCs) such as macrophages. APCs stimulate T lymphocytes and lead to the proliferation of tumor antigen-specific cytotoxic Tcells (CTLs), which then migration into the tumor mass to exert antitumor immune response. In addition, IR promotes proinflammatory macrophages activation and enhances their immune-stimulatory and tumoricidal activities. Macrophages also play a role in attracting innate immune cells, such as NK cells, tumor-associated neutrophils, and probably other types of immune cells. However, in patients with hypertriglyceridemia, it has possibility that the effects of IR on immunogenic cell death are different. Hypertriglyceridemia, which results from elevated Triglyceride (TG) levels in serum, leads to the development of inflammatory response and immune cell death. In previously, several studies have shown that elevated serum levels of TGs increase cell-mediated immune response while reducing the number of immune cells. Therefore, it has possibility that TG activates leukocytes to enhance the immune response including inflammation in the short period, but number of leukocytes is reduced in the long period, leading to immune deficiency.Patients with hypertriglyceridemia need to be further investigated in relation to IR treatment, because the response of immune cells affected by concentration of TGs. We will use the ‘hyper-triglyceride cell model’ to study how IR affects immune cells exposed to high concentration of TGs.
