Triglyceride Induces Eexpression Level of TNF-α and TNF-αR in Jurkat T cells

Triglyceride (TG) is known to be associated with chronic vascular inflammation, and leads to development of atherosclerosis when the condition becomes intensified. In a variety of atherosclerosis models, Tlymphocytes are localized in the earliest lesions of atherosclerosis. T cell associated cytokines such as TNF-α and IFN-γ have pre-dominant inflammatory effects in chronic vascular diseases. To elucidate whether TG affects the expression of inflammation associated cytokines, the mRNA expression level of TNF-α, IFN-γ, IL-1β and TGF- β was analyzed after TG treatment in Jurkat T cells. In this study, we found that TG increases the mRNA expression level of TNF-α in human T cell line dose dependently (0, 0.05, 0.1, 0.2, 0.5, and 1 mg/mL). In a time course experiment, TG (0.5 mg/mL) increases expression of TNF-α beginning at 3h of TG treatment. In contrast, expressions of other cytokines were not changed significantly, regardless of the TG treatment. In addition, it was confirmed that the expression level of TNF-αR also increased by TG in Jurkat T cells. Based on this observation, we suggest that concentration of TG might induce the expression of TNF-α and TNF-αR in Jurkat T cells. Additionally, TG is expected to exacerbate the inflammatory response in chronic vascular diseases.
