Cigarette smoke is one of the most important environmental pollutants that cause DNA damage. FOXO is a subgroup of the forkhead family transcription factor that has a conserved DNA binding site in promotor of its target genes. FOXO has been reported to be involved in various biological functions such as cell cycle control, apoptosis, DNA repair and ROS detoxification though regulation of its target genes. In this study, we investigated whether FOXO3a activation by purple corn extract can contribute to protect from cigarette smoke-induced DNA damage. 50 cigarette smokers were selected by several criteria after testing health state. 25 of them were administered with 2 capsules (300mg/capsule) of purple corn extract and 25 of them were administered with 2 capsules (300mg/capsule) of control (crystalline cellulose) every morning and evening for 4 weeks in Hallym University Chuncheon Sacred Heart Hospital. Biochemical examinations of blood samples of 0, 2, 4 weeks were performed. Cigarette smokers administered with capsule of purple corn extract and with capsule of control did not show any statistical difference in blood levels of ALT and AST (as liver damage markers) and TG. The level of γ-H2AX of blood cells without red blood cells, a DNA damage marker, was decreased in smokers administered with capsules of purple corn extract when compared to smokers administered with control capsules in 2 and 4 weeks. Protein levels of p-AMPK, FOXO3a, p-FOXO3a (thr413), and MnSOD were increased in smokers administered with capsules of purple corn extract when compared to smokers administered with control capsules in 2 and 4 weeks. The result shows that administration of purple corn extract activates p-AMPK and thereby activates p-FOXO3a-413. This activated eventually a FOXO3a target gene, MnSOD. This activation of MnSOD may protect blood cells from cigarette smoke-induced DNA damage.
