영문 초록

The study on the combination of an individual s genetic information with environmental factors (gene-environment interaction) is of great importance to understand the development of diseases, especially lethal diseases including cancer. Several findings demonstrate that the change in expression and activity as well as genetic variability of DNA repair machineries caused by environmental chemical (e.g., heavy metals and cigarette smoke) and physical carcinogens (ultraviolet and ionizing radiation) is likely associated with various diseases. In fact, our group has intensively investigated major DNA repair mechanisms, nucleotide excision repair and bas excision repair, as a crucial pathway in protection of the DNA integrity against environmental toxicants. Also, our in vitro and in vivo studies had identified the effects of antioxidant on modulating the redox and DNA repair system which were vital mechanism in aging. Moreover, the data set of normal people from 1000 genomes database has been utilized to analysis of the property in aging, through a comparative study of genomic variations among populations. Indeed, allele frequency of genetic variation was shown unique properties of DNA repair mechanism in each population who were in different environment. Furthermore, specific diseases or biological processes, which are regulated by major gene set, were predicted by in silico pathway analysis and integrative study on gene-environment interaction using big data database. According to our data, we have suggested a novel insight into further integrative study on aging using genomic data. Additionally, it might be useful to establish a personalized healthcare solution in aging.
