사방오리(Alnus firma) 잎 추출방법에 따른 항균, 항염, 항산화 효과에 관한 연구

국문 초록

본 연구에서는 화장품 원료 소재 개발을 위해 제주에서 자생하는 사방오리 잎의 열수추출물과 에탄올추출물, 그리고 Lactiplantibacillus plantarum로 발효한 사방오리 잎의 에탄올 추출물에 대한 항균, 항염 및 항산화 효과에 대한 비교분석을 수행하였다. 대표적인 피부 유해균인 Staphylococcus aureus, Pseudomonas aeruginosa, Corynebacterium diphtheriae, Streptococcus pyogenes, Acinetobacter junii, Cutibacterium acnes 및 Stenotrophomonas maltophilia에 대하여 디스크확산법을 통하여 항균효과를 검증하였고, 항염증 효과 확인을 위해 NO 생성 억제 실험, 항산화 효과를 위해서는 DPPH assay와 ABTS assay를 수행하였다. 실험 결과 사방오리 잎 열수추출물과 발효물에서는 항균 효과는 미미하였으나, 에탄올추출물에서는 C. acnes에서 가장 뛰어난 항균 효과를 보였으며, A. junii, S. maltophilia에서도 우수한 항균효과를 보였다. 항염증 실험 결과 열수와 에탄올추출물에서는 항염증 효과가 미미하였으나, 발효물에서 항염증 효과를 보였으며 항산화 효과는 열수추출물, 에탄올추출물 및 발효물 모두에서 높은 항산화 효과를 확인 할 수 있었다. 이러한 결과는 제주에서 자생하는 사방오리가 피부 건강 제품 개발에 좋은 소재로 활용될 수 있음을 시사하며, 특히 에탄올추출물은 항균작용 및 항산화 효과가 있는 화장품 원료로 뛰어난 가치가 있음을 보여준다.

영문 초록

This study explores the antibacterial, anti-inflammatory, and antioxidant properties of hot water extract, ethanol extract, and fermented products derived from the leaves of Alnus firma, a plant native to Jeju Island, with the aim of developing materials for skin health applications. Antibacterial activity was assessed against skin-harmful pathogenic bacteria, including Staphylococcus aureus, Pseudomonas aeruginosa, Corynebacterium diphtheriae, Streptococcus pyogenes, Acinetobacter junii, Cutibacterium acnes, and Stenotrophomonas maltophilia, using the disk diffusion method. Anti-inflammatory effects were evaluated through nitric oxide (NO) production inhibition assays, while antioxidant activities were measured using DPPH and ABTS assays. Among the tested samples, the ethanol extract exhibited the strongest antibacterial activity against C. acnes, A. junii, and S. maltophilia. Although the hot water and ethanol extracts showed limited anti-inflammatory effects, the fermented product demonstrated significant activity. All three samples exhibited high antioxidant activity. These findings suggest that A. firma leaves hold significant potential as effective ingredients for skin health products. Notably, the ethanol extract stands out as a promising candidate for cosmetic applications due to its potent antibacterial and antioxidant properties.
