국문 초록
흰쥐 대뇌 변연계의 몇몇 부위에서 강박 스트레스로 인한 Fos 발현 정도와 diazepam의 효과를 평가하여
스트레스에 의한 뇌의 Fos 발현이 diazepam의 영향을 받는지를 알아봄으로써 diazepam의 항불안효과와
관련된 뇌 부위를 규명하고자 본 실험을 시도하였다.
흰쥐 뇌에서 강박 스트레스와 diazepam에 의한 Fos 발현의 차이를 Fos 항체를 이용한 면역조직화학법으
로 관찰하였다. 실험 동물로는 체중 200∼250g의 Sprague-Dawley 수컷 흰쥐 20마리를 사용하였으며 4
개군으로 나누어 각군마다 5마리씩을 배당하였다. 실험군은 생리식염수를 투여한 후 강박 스트레스를 가한
군, diazepam(5mg/kg)을 투여한 후 강박 스트레스를 가한 군, diazepam(5mg/kg)만을 투여하고 강박 스트
레스를 가하지 않은 군, 그리고 대조군으로 생리식염수 1 ml만을 투여하고 강박 스트레스를 가하지 않은 군
등 모두 4개군이었다.
뇌 부위별로 c-fos 단백질 면역반응성 세포의 수를 측정하여 다음과 같은 결과를 얻었다.
1) 해마영역 CA2와 치아이랑을 포함하는 해마형성체의 Fos 발현은 강박군에서 다른 세 군에 비하여 뚜
렷이 증가하였다. 또한 각 군의 Fos 발현에 대한 검증에서는 스트레스와 diazepam 모두가 유의한 영향을
미치는 것으로 나타났으며 두 요인의 교호작용도 유의한 영향을 나타냈다.
2) 편도핵의 편도해마영역에서 강박군의 Fos 발현은 diazepam군에 비하여 유의한 차이를 나타냈으며 대
조군과 diazepam-강박군에 비하여 유의한 차이가 없었다. 각 군의 Fos 발현에 대하여 스트레스와 diazepam
모두가 유의한 영향을 미치는 것으로 나타났으며 두 요인의 교호작용은 유의하지 않았다.
3) 뇌실옆핵에서 강박군의 Fos 발현은 대조군과 diazepam군에 비하여 유의한 차이를 나타냈으며 dia-
zepam-강박군에 비하여 유의한 차이가 없었다. 각 군의 Fos 발현에 대하여 스트레스만이 유의한 영향을
미치는 것으로 나타났으며 스트레스와 diazepam 두 요인의 교호작용도 없었다.
4) 전외측슬상핵에서 강박군의 Fos 발현은 diazepam군과 diazepam-강박군에 비하여 유의한 차이를 나
타냈으며 대조군에 비하여 유의한 차이가 없었다. 각 군의 Fos 발현에 대하여 diazepam만이 유의한 영향을
미치는 것으로 나타났으며 스트레스와 diazepam 두 요인의 교호작용도 없었다.
5) 유두체상핵, 측두피질영역 3, 후두피질영역 2, 이상피질, 그리고 대상피질영역 3에서는 각 군간에 Fos
발현이 유의한 차이가 없었고 각 군 모두에서 Fos 발현이 스트레스에 의하여 통계적으로 유의한 영향을 받
지 않았으며 diazepam에 의해서도 유의한 영향을 받지 않았다.
본 연구를 통하여 스트레스와 diazepam이 모두 해마형성체에서 Fos 발현에 유의하게 영향을 준다는 사실
을 알 수 있었다. 이는 diazepam의 항불안작용이 해마형성체를 통하여 조절된다는 추측을 가능케 하고 해마
형성체에서 스트레스 후 증가되었던 Fos 발현이 diazepam에 의한 항불안작용의 결과로 감소된 것으로 판단
된다.
영문 초록
This study was conducted to identify the effects of diazepam on stress-induced c-fos expression in
rat brain.
The rats were divided into 4 groups according to injection agents [0.9% isotonic saline 1ml,
diazepam(5 mg/kg)] and restraint stress, respectively. Saline or diazepam was injected into the
peritoneum of rats 10 minutes before the restraint groups were restrained in a wire chamber.
Fos-immunoreactive neurons were calculated in the hippocampal area CA2, dentate gyrus, amygdalohippocampal
area, paraventricular nucleus, ventral lateral geniculate nucleus, supramammillary
nucleus, temporal cortex area 3, occipital cortex area 2, piriform cortex, and cingulate cortex area 3.
The results were as follows:
1) In the hippocampal formation including hippocampal area CA2 and dentate gyrus, Fos
expression increased significantly in the saline-injected restraint animals than in any other groups. A
two-way factorial ANOVA revealed a significant effects of stress and diazepam, and a significant
stress by diazepam interaction.
2) In amygdalohippocampal area, Fos expression increased significantly in the saline-injected
restraint animals than in diazepam-injected animals. A two-way factorial ANOVA revealed a significant
effects of stress and diazepam, but no significant interaction of stress and diazepam.
3) In paraventricular nucleus, Fos expression increased significantly in the saline-injected restraint
animals than in saline-injected animals and diazepam-injected animals. A two-way factorial ANOVA
revealed a significant effects of stress, no significant effects of diazepam, and no significant interaction
of stress and diazepam.
4) In ventral lateral geniculate nucleus, Fos expression increased significantly in the saline-injected
restraint animals than in diazepam-injected animals and diazepam-injected restraint animals. A twoway
factorial ANOVA revealed a significant effects of diazepam, no significant effects of stress, and
no significant interaction of stress and diazepam.
5) In supramammillary nucleus, temporal cortex area 3, occipital cortex area 2, piriform cortex, and
cingulate cortex area 3, there was no significant difference of Fos expression in each group. A twoway
factorial ANOVA revealed no significant effects of stress and diazepam, and no significant
interaction of stress and diazepam. These results suggest that the hippocampal formation is involved
in the response to the stress and the tension reduction effect of diazepam.
