C57BL-6형 백서의 알코올 섭취량에 대한 Naltrexone과 Olanzapine의 효과

국문 초록

연구목적： 본 연구는 현재 임상에서 알코올리즘 치료에 사용중인 오피오이드 길항제인 naltrexone과 세로토 닌-도파민 수용체 길항제인 olanzapine를 투여한 후 알코올 섭취를 강화시킨 C57BL-6형 백서의 알 코올 섭취량 변화를 비교하였다. 방 법： C57BL-6형 백서 28마리에게 30일 동안 하루에 2시간씩 소량의 에탄올과 물이 제공되었다. 31일 째부터는 백서를 네 군으로 나누어 음주 30분전에 각기 다른 종류의 약물과 위약을 복강내 투여했다. 그후 35일 동안 약물 투여군에는 각각 naltrexone 5mg/kg, olanzapine 0.1mg/kg, olanzapine 1.0 mg/kg을, 대조군에는 증류수를 매일 투여했다. 결 과： 대조군과 비교하여 naltrexone 5mg/kg을 투여한 군은 백서의 알코올 섭취량이 4주째부터 유의하게 감소되었고, olanzapine 0.1mg/kg을 투여한 군은 약물투여 2주 째부터 5주 째까지 섭취량이 감소하 였으며, olanzapine 1mg/kg에서는 섭취량이 2, 3주째 감소하였으나 그 후로는 알코올 섭취량이 유의 하게 감소되지 않았다. 알코올 섭취를 강화시킨 백서에서, 알코올 섭취량을 감소시키는데 olanzapine 1mg/kg보다는 0.1mg/kg이 효과가 있었다. 그러나 olanzapine 0.1mg/kg이 naltrexone 5mg/kg 보 다 유의하게 알코올 섭취를 감소시키지는 않았다. 결 론： Naltrexone 뿐만 아니라 적은 용량의 olanzapine에서도 C57BL-6형 백서에게 단기간의 알코올 섭 취량의 감소가 있었다

영문 초록

Objectives：This study compared the effects of opioids antagonist naltrexone and serotonin-dopamine recepter antagonist olanzapine on the reduction of alcohol consumption level of alcohol intake reinforced C57BL-6 type rat. Methods：Small amount of alcohol and water were applied to the 28 rats for 2 hours per day during 30 days. On the 31th day, The rats were divided into four groups and given different medications by intraabdominal route 30 minutes before the alcohol consumption. For the next 35 days, the 3 subject group were applied with naltrexone 5mg/kg, olanzapine 0.1mg/kg and olanzapine 1.0mg/kg and the control group with distilled water everyday. Results：In contrast to control group, naltrexone 5mg/kg group showed a significant reduction of alcohol consumption after 4 weeks. Olanzapine 0.1mg/kg group showed a decrease of alcohol consumption from 2 to 5 week period. Although olanzapine 1mg/kg group showed a momentary decrease of the consumption during the 2nd and 3rd weeks, the group did not show significant decrease afterwards. Olanzapine 0.1mg/kg was more effective in reducing the alcohol consumption than olanzapine 1mg/kg. However, it is not significantly more effective compared to the naltrexone 5mg/kg in reducing the alcohol consumption of the reinforced rats’ alcohol intake. Conclusion：This results suggest that the low dose of olanzapine as well as naltrexone reduce the alcohol intake in C57BL-6 type rats.
