Protective Effects of Cuscutae Semen Extract from Oxidative Stress and Amyloid β25-35-Induced SH-SY5Y Neuronal Cell Dysfunction

Objectives: Amyloid beta (Aβ) is generated by the amyloidogenic pathway. Its accumulation is a major cause of Alzheimer’s disease. The present study investigates the free radical scavenging activities and inhibition of Aβ25-35-induced neurotoxicity by Cuscutae semen (CS) water extract treatment. Methods: The scavenging activities of radicals, including 2, 2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl (OH), and nitric oxide (NO) of the CS water extract were assessed. Moreover, the SH-SY5Y cells were used to investigate cell viability, inflammation-related protein expression, and inflammatory cytokine secretion using CS water extract to determine neuroprotective effects. Results: The CS water extract inhibited the production of free radicals such as DPPH, OH, and NO, dose- dependently. Meanwhile, oxidative stress was induced, and cell viability was decreased by Aβ25-35. The CS water extract treatment significantly increased cell viability in Aβ25-35-stimulated SH-SY5Y cells. Additionally, the treatment with CS water extract significantly down-regulated the expression of inducible nitric oxide synthase and cyclooxygenase-2 in Aβ25-35‐stimulated SH-SY5Y cells. Secretion of interleukin-1β and interleukin-6 in Aβ25-35‐induced SH-SY5Y cells was significantly inhibited by CS water extract. Conclusions: This study indicated the free radical scavenging activities and neuroprotective effects of CS water extract.
