국문 초록

연구목적：본 연구는 예측불가능한 경도의 만성적 스트레스를 백서에게 가하여 우울 상태에 준하는 상태 를 유발한 후 sertraline을 투여하여 1% 자당용액 섭취량과 체중에 미치는 영향을 정상백서와 비교하고자 하였다. 방 법：11가지 종류의 스트레스를 18마리의 백서에게 4주간에 걸쳐서 처리하여 우울상태에 준하는 자당 섭취량의 저하를 확인하였다. 스트레스로 처리된 실험집단을 무작위로 sertraline 투여군(N＝9)과 saline 투여 군(N＝9)으로 나누었으며 스트레스 처리를 하지 않은 18마리의 통제집단도 sertraline 투여군(N＝9)과 saline 투여군(N＝9)으로 나누었다. 실험집단과 통제집단은 이후 4주간에 걸쳐서 sertraline(4.29mg/Kg) 또는 saline( 0.5cc)을 매일 복강내 주사하였다. 실험집단은 실험약물 주사 기간중 지속적으로 스트레스 처리를 하였 다. 실험이 진행되는 전체기간중 매주마다 1% 자당섭취량과 체중을 8주간에 걸쳐서 측정하였다. 실험결과：1) 실험집단의 sertraline 투여군은 실험 1주와 4주사이에 자당섭취량의 유의한 증가가 있었다. 통제집단의 sertraline 투여군은 자당섭취량이 저하되는 경향을 보였으나 통계적으로 유의하지는 않았다. 2) 실험집단의 sertraline 투여군은 저하된 체중이 그대로 유지되었으며 통제집단의 sertraline 투여군은 실험 1주 와 3주, 4주 사이에 유의한 체중의 증가를 보였다. 3) 실험집단과 통제집단은 자당섭취량과 체중사이에 유의한 상관을 보이지는 않았다. 결 론：Sertraline은 만성적으로 예측불가능한 경도의 만성적 스트레스를 가한 백서에서 저하된 1% 자당섭 취량을 원래상태로 회복시키는 소견을 보였으나 체중은 회복시키지 못하였다. 정상백서에서 sertraline은 1% 자당 섭취량의 저하를 유발하였으나 체중에는 영향을 미치지 않았다.

영문 초록

The study was designed to evaluate the effects of sertraline on 1% sucrose intake and weight change in rats with chronic unpredictable mild stress and normal controls. We applied 11 types of stress regimens and identified depressive behaviours in 18 Spraque-Dawley rats for 8 weeks. After 4 weeks of chronic unpredictable mild stress procedure, those 18 rats were stratified into a sertraline-treated sabgroup and a saline sabgroup. Also nonstressed 18 rats were stratified into the sertraline-treated sabgroup and the saline sabgroup and were started intraperitoneal injections of sertraline(4.29mg/Kg) or saline for rest of 4 weeks. The 1% sucrose intake and the body weight were checked on the 4th day of every week, over the 8 weeks of experiment. The results were as follows： 1). The sertraline-treated sabgroup of chronic unpredictable mild stressed rats showed significant increase of 1% sucrose intake between the 1st week and the 2nd, the 3rd and 4th week, while the sertraline- treated sabgroup of non-stressed rats showed decreasing trend for 1% sucrose intake. 2) The sertraline-treated sabgroup of chronic unpredictable mild-stressed rats showed a sustained decrease of body weight, while the sertraline-treated sabgroup of non-stressed rats showed a non-significant increase of body weight. 3) In the group subjected to chronic unpredictable mild stress, there were no significant correlations between 1% sucrose intake and body weight and also no correlations in the non-stressed group. In summary, sertraline had an effect on restoring the decreased 1% sucrose intake to normal condition but no effect on regaining the body weight in the chronic unpredictable mild stress-treated rats. Sertraline resulted in a decrease of 1% sucrose intake and no effect on body weight in the non-stressed rats.
