항정신병약물이 백서뇌 c-fos 및 jun B 의 발현에 미치는 영향

영문 초록

To study the action mechanism of antipsychotics in the molecular level, we observed the changes in the induction of c-fos and ju n B genes in rat brain by chlorpromazine and haloperidol through Northern blot analysis. We could observe the induction of c-fos and ju n B in striatum, cerebral cortex and hippocampus by chlorpromazine. But we could not detect definite differences between experimental and control groups(injected with water) in cerebral cortex and hippocampus. However in the striatum, increases of c-fos and ju n B m R N A in experimental groups were more than those in sham groups and were dependent on dose of chlorpromazine. The inductions of c-fos and ju n B m R N A increased initially at 15min, reached peak level at 45min and returned to basal level around 90min. We observed the similar results from haloperidol dissolved with tartaric acid. Definite differences between experimental and sham groups could be detected in only striatum. From these results, we know that chlorpromazine induce c-fos in rat striatum like haloperidol and ju n B also induced by antipsychotics in similar patterns of c-fos. We also observed nonspecific induction of these genes in other brain regions except for the striatum. We suggest that these result from nonspecific effects like the irritation caused by intraperitoneal injection and solvent But there are still possibilities that the specific inductions by drugs are masked by these nonspecific effect
