영문 초록

In this study, it was investigated to be able to use direct polymerase chain reaction (PCR) for rapid detection of Listeria monocytogenes in chickens. A pair of primer based on a unique region in the 16S rRNA sequence of L. monocytogenes was used to perform the direct PCR and to examine its sensitivity. The trial was based on sampling of the chicken meat followed by swabbing and rinsing methods and centrifugation. As a result of this procedure, the inhibitor of DNA amplication was excluded and concentration of L. monocytogenes was condensed more. The direct PCR could detect as few as 4 cells/cm² by swabbing and 40 cells/g by rinsing. When compared these methods with culture method for detection of L. monocytogenes in chicken meat, the swabbing and the rinsing method was, respectively, 88.6% and 60.0% agreememt with culture method. However, a result of the direct PCR using swabbing method agreed to that of culture method in all trial tested adding 10 ¹cells/cm². It indicates that the sensitivity of the direct PCR can be increased by swabbing method. In this procedure, analysing time, including sample treatment steps were not more than 6 h.
