Pasteurella multocida, an organism commonly found in immunosuppressed pigs due to an infection with Mycoplasma hyopneumoniae were experimentally inoculated into pigs that were previously infected with M. hyopneumoniae, and the pathological interaction of the two organisms in the formation of the pneumonic lesions, distribution of the organisms and the lymphocyte infiltration in the pulmonary tissues were investigated. Changes in leukocyte subpopulation in the peripheral blood during the course of pneumonia were monitored using flow cytometry from which the pathological contribution of the two organisms to the pneumonic lesions and the local/systemic immune responses were evaluated.
Eleven one-month-old male pigs which were respiratory pathogen-free and seronegative to M. hyopneumoniae and P. multocida were divided into two group : six pigs in the test group were inoculated with inoculation 4×10⁻⁷ CFU /ml M. hyopneumoniae J101 strain in 4ml once every two days for 9 weeks, followed by 6 inoculations of PHLS strain by the same protocol. Five pigs were served as uninfected control group. One pig in the test group was euthanized to confirm the experimental infection of M. hyopneumonia, by macroscopic, histopathologic and immunohistochemical examinations. Pigs were subsequently inoculated with same dose of 10ml of 6×10⁻⁶ CFU/ml A and D type via the intratracheal route and were euthanized at 1, 3, 7, 14 and 21 days after infection.
Macroscopic pneumonic lesions caused by the double infection with M. hyopneumoniae and P. multocida included dark brownish consolidated multifocal lobular lesions, moderate multifocal hemorrhages and severe fibrosuppurative exudate in the bronchi and bronchioli at day 1 after inoculation with P. multocida. The pneumonic lesions were most extensive at 3 days after inoculation with P. multocida, after which lesions began to subside. Histopahological changes included fibrosuppurative lesions in bronchi and bronnchioles to the degree similar to that of macroscopic observation. A moderate lymphocyte infiltraion around bronchi, bronchioles and blood vessels were observed throughout the course of post inoculation with P. multocida.
The immunoperoxidase staining of M. hyopneumoniae and P. multocida, in the pulmonary tissues demonstrated variable numbers of M. hyopneumoniae organisms in the bronchi, bronchioles, alveoli tissues and on their cilia. On the contrary, organisms of P. multocida were observed in or around inflammatory cells, especially neutrophils, red blood cells and macrophages infiltraed around pustular lesions on 1, 3, and 14 days after inoculation. Clumps of organisms floating inside the alveoli were also observed. Pigs subsequently infected with P. multocida follwed by the exposure of M. hyopneumoniae, demonstrated an increased infiltration of B cells in the pulmonary tissues using by immunohistochemistry, whereas a decreased expression of CD45, CDlla and increased MHC class Ⅱ antigens on the peripheral blood leukocytes using by flow cytometry were observed.
Data obtained from this study indicated that M. hyopneumoniae elicited a significantly fibrosuppurative bronchopneumonia in pigs superinfected with P. multocida. The immunohistochemical and flow cytometric analysis of local and systemic immune response of pig showed that both local and systemic humoral immune responses were enhanced in pigs double-infected with M. hyopneumoniae and P. multocida.
