국문 초록

동물대체시험법은 21세기 산업과 환경의 지속 가능한 발전을 위한 중요한 목표 중 하나로 부각되고 있다. 이러한 대체시험법은 화학물질, 의약품, 화장품 등의 안전성과 유효성을 평가함에 있어 동물과 인간의 종간 차이를 극복하고, 동물 실험의 윤리적, 경제적 문제를 해결하고 정밀의료를 실현하고자 하는 현대 사회의 요구에 부응한다. 미국, 유럽, 일본 등 주요 선진국과 OECD, ISO와 같은 국제 표준 개발 기구들은 이러한 사회적 요구에 부응하기 위해 노력하고 있다. 특히, 오가노이드를 활용한 대체시험법은 인간의 장기나 조직을 모사한 3차원 세포 배양 기술로서, 실제 인간에 대한 반응을 정확하게 예측할 수 있는 장점이 있어 주목받고 있다. 이 기술은 동물실험의 윤리적 문제를 해결할 뿐만 아니라, 실험 결과의 정확도를 향상시킬 수 있는 혁신적인 방법으로 대두되고 있다. 본 논문은 오가노이드를 활용한 대체시험법의 선도적 국제 표준 개발을 목적으로 관련 국내외 표준현황을 포괄적으로 분석한다. 주요 국가의 대체시험법과 규제 정책 및 제도를 조사하고, 오가노이드 기반 대체시험법의 국내외 표준화 진행 현황을 검토하며, 오가노이드의 장기별 개발 프로토콜을 분석한다. 또한 동물대체시험법의 표준화가 나아갈 방향에 대한 전망을 제공하고, 이 분야에서의 기술적, 규제적 도전과제를 탐색한다. 연구 결과, 오가노이드 기반 대체시험법은 국가별 규제 정책에서 동물실험을 대신하는 비임상시험으로 인정되기 시작했지만, OECD, ISO에서 표준개발은 시작 단계이다. 간, 장, 심장 등 장기별 오가노이드 제작 프로토콜 개발은 급증하고 있고 필수 요건에 대한 이해관계자들의 합의가 마련되지 않아, 국제적인 협력은 표준화를 앞당기고 오가노이드 기반 대체시험법의 상용화를 촉진할 것으로 예상된다. 대체시험법 표준화 현황, 도전 요인 및 미래 전망을 본 논문에 상세히 제시하여, 지속 가능한 발전과 동물 보호에 기여할 수 있는 대안적인 방법들을 모색하는 데 크게 이바지할 수 있을 것으로 기대되며 본 연구가 동물대체시험법의 발전뿐만 아니라, 인간 중심의 정밀한 안전성 평가에 있어 중요한 전환점을 마련할 것으로 보인다.

영문 초록

Animal testing for toxicity assessments has long been a point of contention due to concerns over animal welfare and ethics. As an integral component of product development and consumer safety, the evaluation of chemical toxicity is vital. Yet, the 21st century has witnessed a growing demand for alternative testing methods that can offer sustainable solutions without relying on animal experiments. This need has spurred major developed countries and international standardization organizations to pursue the development and standardization of novel testing approaches. Among these, organoid-based methods have emerged as a promising avenue for innovation. This study examines the concept of organoids, defining and categorizing alternative testing methods. It explores a broad spectrum of topics, including the standardization trends of alternative testing method regulations, organoid-based method standardization, and trends led by government entities, research institutions, academia, and corporations. It also examines the development of organ-specific manufacturing protocols for organoids. The research finds that international efforts to standardize alternative testing methods are in progress, with a particular focus on organoid-based strategies. The anticipated growth and evolution of such alternatives could revolutionize standard practices in safety assessments and drug efficacy evaluations. The standardization of organoid-based testing methods is poised to become a crucial milestone in advancing human health and environmental protection, driving the sector's growth and innovation. Moreover, advancements in organoid technology and the creation of models for various organs are likely to further refine alternative testing methods and introduce new evaluation techniques for drugs and chemicals. This study highlights to the biomedical community that standardizing new testing methods can address the ethical dilemmas of animal testing, underscoring a significant contribution towards ethical scientific practices.
