영문 초록

This study was conducted to induce hairy roots from indigo plant and select proper Agrobacterium rhizogenes out of seven strains for production of indigo pigment in hairy root culture of Polygonum tinctorium. In general, hairy root formation on leaf was better than on stem in all Agrobacterium rhizogenes strains used. MS was the best medium followed by White and B₅ media in order for hairy root induction. Strains 15834 and A₄ induced hairy root more effectively from leaf on MS medium. All strains grew well on MS rather than on White or B₅ media. Hairy roots induced by strains 15834 and A₄ on MS medium showed rapid growth compared with those induced by other strains. Indigo content was the highest in strain 11325 on MS medium even though the growth rate was the lowest among strains used. Strains 15834 and A₄ were selected for production of indigo by hairy root culture based on hairy root induction, growth rate and indigo accumulation in the hairy roots. Opine was detected in the induced hairy roots indicating the occurence of transformation from A. rhizogenes.
