국문 초록
목 적:Fas 변이를 보이는 lpr 마우스와 Fas ligand 변이를 보이는 gld 마우스를 이용하여 in vivo에서 Fas와 Fas ligand의 발현이 전리 방사선에 의해 유도되는 apoptosis에서 어떤 역할을 하는지 조사하고자 하였다.
대상 및 방법 : Fas의 변이를 보이는 C57BL/6J-Faslpr 마우스와 대조군인 C57BL/6J 마우스, Fas ligand 변이를 보이는 C3H/HeJ- Fasgld 마우스와 대조군인 C3H/HeJ 마우스를 대상으로 하였다.마우스는8주령 웅성으로서 전신 방사선 조사하여 일정 시간 후 비장을 적출하였다. 조직을 hematoxylin-eosin 염색하여 apoptosis 유도 수준을 비교 분석하였다.또한 apoptosis 조절 물질인 p53,Bcl-2, Bax, Bcl-XL,Bcl-X S에 대하여 Westernblotting을 시행하고 발현수준을 densitometry로 분석하여 관련된 기전을 연구하였다.
결 과: 2.5 Gy와 10 Gy 조사시에 C57BL/6J-Faslpr 마우스와 C3H/HeJ-Fasgld 마우스에서 대조군에 비하여 방사선에 의한 apoptosis가 유의하게 감소되는 것으로 나타났다(p<0.05).C57BL/6J 마우스와, C3H/HeJ 마우스에서 10 Gy 방사선 조사후 Bax가 8시간째에 각각 3배, 3.3배의 증가를 보였으나 C57BL/6J-Faslpr 마우스와,C3H/HeJ- Fasgld 마우스에서는 뚜렷한 발현증가가 관찰되지 않았다.
결 과: Fas의 변이가 있는 lpr 마우스와 Fas ligand의 변이가 있는 gld 마우스에서 방사선에 의한 apoptosis가 대조군 보다 현저하게 낮으며 이는 방사선에 의한 Bax의 유도가 미약한 것과 연관된 것으로 나타났다.방사선에 의한 apoptosis 유도에 Fas의 역할이 매우 중요한 것으로 보인다.
영문 초록
Purpose: The interaction of the Fas: Fas ligand has been recognized to play an important role in radia-tion-induced apoptosis. The purpose of this study was to investigate the role of Fas and Fas ligand mutations, in radiation-induced apoptosis in vivo.
Materials and Methods Mice with a mutation in the Fas (C57BL/6J-Faslpr) and its normal control (C57BL/ 6J) and the Fas ligand (C3H/HeJ-F a sgld) and its normal control (C3H/HeJ), were used in this study. Eight-week old male mice were given whole body radiation. After irradiation, the mice were killed at various time intervals, and the irspleens collected. Tissue sample was stained with hematoxylin-eosin, and the number so fapoptotic cells scored. The regulating molecules of apoptosis including the p53, Bcl-2, Bax, Bcl-X L and Bcl-X S genes were also analyzed by Western blotting.
Results: With 2.5 Gy and 10 Gy of irradiation, the levels of apoptosis were lower in the C57BL/6J-Faslpr and
C3H/HeJ-Fasgld mice than in the control mice (p 0.05). With the expression of apoptosis regulating molecules, the B ax was increased in both the C57BL/6J and C3H/HeJ mice in response to radiation; the peak levels o f B ax in the C57BL/6J and C3H/HeJ were 3 and 3.3-fold higher after 8hr, respectively. However the Bax was not increas ed in either the C57BL/6J-Faslpr or C3H/HeJ-Fasgldmice. The p53, Bcl-X L, Bcl-X S and Bcl-2 showed no significant changes in the C57BL/6J-Faslpr, C 3 H / H e J - F a sgld, C57BL/6J and C3H/HeJ mice.
Conclusion The levels of radiation-induced apoptosis w e re lower in the lpr and gld, than the control mice,
which seemed to be related to the level o f B ax activation due to the radiation in the lpr and gld mice. This
result suggests that Fas/Fas L plays an important role in radiation-induced apoptosis in vivo .
