This research was conducted to find the possible controlling mechanism of the extraordinary stem elongation of soybean breeding line Super Tall which was a F₆ offspring derived from a cross of Flambeau and Shinsei. The phenotype of Super Tall closely resembled to soybean plants treated with exogenous gibberellic acid(GA). The involvement of GA in Super Tall stem growth was studied. GA was extracted from 15-day-old soybean plants, partly purified, and subjected to thin layer chromatography. Gibberellin activity was assayed with dwarf rice seedlings. The assay demonstrated that Super Tall contained higher level of gibberellin and lower level of inhibitors than its parent cultivars did. To clarify the cause of drastic stem elongation of Super Tall was the result either of high efficiency of GA production and turnover, or of high sensitivity of soybean to GA, plant response to the application of exogenous GA₃ and GA biosynthesis inhibitor, AMO-1618 were measured. Elevated temperature enhanced the shoot growth and increased fresh weight of soybeans. Temperature affected responsiveness of soybean to exogenous GA₃ treatmant. Application of exogenous GA₃ was less effective at high temperature than at low temperature. The inhibitory effect of AMO-1618 on stem elongation was more apparent at high temperature(27°/21℃) than at low temperature(21°/15℃). This suggest that the endogenous level of GA was higher at high temperature than at low temperature. The stimulatory effect of GA₃ on stem growth of soybeans was much greater in Short Sister Selection than in Super Tall, and the inhibitory effect of AMO-1618 on the soybean growth was more distinctive in Super Tall than in Short Sister Selection. This may be interpreted as the result of the higher level of endogenous gibberellin in Super Tall than in Short Sister Selection, but not the result of the higher sensitivity of Super Tall to gibberellin. Abscisic acid(ABA) was extracted, partially purified, and seperated by thin layer chromatography. The ABA activity was assayed by wheat coleoptile elongation in the dark. The level of ABA was high in short parent cultivar Flambeau, low in Super Tall, and medium in Shinsei. In conclusion, it is proposed that the enhanced stem elongation of Super Tall was the result of the combination of high level of endogenous GA and low level of ABA.
