Jak2 억제제인 AG490이 뼈파괴세포 (Osteoclast)의 분화에 미치는 영향

국문 초록

뼈파괴세포는 혈구형성세포에서 기원된 세포로 뼈 흡수에 중요한 역할을 하고 뼈엉성증(Osteoporosis), 치주염, 류마티스 관절염 같은 뼈 질환의 원인이 된다. 본 연구에서 우리는 뼈파괴세포의 분화에 Jak2 특이적 억제제인 AG490의 영향을 검증하였다. AG490은 유의하게 농도 의존적으로 Receptor activator of NF-κB ligand (RANKL)에 의해 매개되는 뼈파괴세포의 분화를 억제하였다. 또한, RANKL은 p38, ERK, JNK의 인산화를 촉진하였고 I-κB의 소멸을 유도 하였으나 AG490은 RANKL에 의한 p38 인산화를 억제하였다. AG490은 RANKL로 투여한 포식세포에서 발현되는 tartrate-resistant acid phosphatase (TRAP), c-Fos, nuclear factor of activated T cells (NFAT) c1, OSCAR mRNA를 억제하였다. 또한 AG490은 RANKL에 의한 c-Fos와 NFATc1 단백질 발현을 억제하였다. 이들의 결과로 AG490이 c-Fos와 NFATc1 발현을 억제시킴으로써 뼈파괴세포의 분화를 억제한다고 제안한다.

영문 초록

Osteoclasts are cells of hemopoietic origin that play an critical role in bone resorption and responsible for bone diseases, including osteoporosis, periodontal disease, and rheumatoid arthritis. In this study, we examined the effect of AG490, a Jak2-specific inhibitor on osteoclast differentiation. AG490 significantly inhibited receptor activator of NF-κB ligand (RANKL)-mediated osteoclast differentiation in a dose-dependent manner. RANKL stimulated the phosphorylation of p38, ERK, and JNK and promoted I-κB degradation. However, AG490 suppressed the phosphorylation of p38 induced by RANKL treatment. AG490 suppressed the mRNA expression of TRAP, c-Fos, NFATc1, and OSCAR in bone marrow-derived macrophages (BMMs) treated with RANKL. Also, AG490 significantly inhibited the protein expression of c-Fos and NFATc1 in response to RANKL. These results suggest that AG490 inhibited osteoclast differentiation by suppressing the expression of c-Fos and NFATc1.
