돼지 콩팥세관 상피세포에서 occludin의 발현

국문 초록

Occludin은 상피세포의 치밀이음부에 존재하는 주요 접합분자로, 상피세포의 극성을 유지하는 역할을 하고 세포사이를 통한 물질의 이동을 차단하는 역할을 한다. Occludin은 콩팥에서 발현되는 것으로 알려져 있으나 세뇨관에 따른 정확한 발현양상은 아직 알려져 있지 않다. 본 연구의 목적은 콩팥세관에 따른 occludin의 발현양상을 관찰하는 것이다. 성체삼원교잡종(Landrace, Yorkshire, Duroc) 돼지의 콩팥을 분리하여 면역조직화학염색과 immunoblot을 실시하였다. 세뇨관에 따른 occludin의 분포를 관찰하기 위해 세관상피세포의 지표인 aquaporin1 (AQP1), Na+-K+-ATPase β1, thiazide-sensitive Na+/Cl- cotransporter (TSC), calbindin-D28k, H+-ATPase 항체들로 occludin과 함께 이중염색을 실시하였다. 돼지콩팥에서 occludin은 상피세포의 치밀이음부에서 발현되었다. 집합관과 연결세관, 먼쪽세관 상피세포에서 강하게 발현되었고, 헨레고리로 가면서 점점 약하게 나타나는 결과를 보였으며, 토리상피세포와 토리곱슬세관 부분에서는 거의 발현되지 않았다. 발현양상은 세포의 치밀이음부에 점 또는 선의 형태로 발현되었다. 이상의 결과로 occludin은 콩팥세관 상피세포에 따라 다른 발현양상을 보이며, 집합관과 먼쪽세관에서 주요 접합분자로 작용함을 알 수 있었다.

영문 초록

Occludin is a cell adhesion molecule that is abundantly expressed in the kidney. However, the expression pattern in various renal epithelial cells is not well established. The purpose of this study was to determine the cellular localization along the tubular epithelial cells in the kidney. Kidneys from adult pigs crossbred of Yorkshire, Landrace and Duroc (three breeds) were processed for immunohistochemistry. Thiazide sensitive sodium chloride cotransporter (TSC), Na+-KATPase β1, calbindinD28k, and H+-ATPase were used to identify the thick ascending limb, distal convoluted tubule, connecting tubule, and collecting duct, respectively. In the pig kidney, occludin was expressed in the apical domain of the tubular epithelial cells. The immunoreactivity of occludin was strongest in the collecting duct, and then gradually decreased in the connecting tubule, distal convoluted tubule, and thick ascending limb. Occludin expression was weak in the thin limbs of the loop of henle and in the proximal tubule in the pig kidney. These results suggest that occludin may be a major adhesion molecule in distal tubular epithelial cells and play a critical role in maintaining epithelial polarity of these nephron segments.
