Staphylococcus aureus에 대한 한약재의 항균활성

국문 초록

18종의 한약재 MeOH 추출물과 EtOH 추출물의 Staphylococcus aureus에 대한 항균력을 조사한 결과 하고초, 소목, 오배자 및 향유는 5mg/ml 수준에서 항균활성을 나타내었으며 지실, 금은화 및 황금 등은 10mg/ml, 오미자, 초두구 및 희렴 등은 30mg/ml 이상에서 항균성을 보였다. 최소저해농도는 MeOH추출물에서는 1∼34mg/ml, EtOH추출물에서는 1∼46mg/ml 농도에서 항균활성을 보였으며 분획 층 중 EtOAc층에서 성장억제효과가 가장 높게 나타났다. 항균력이 가장 우수한 것으로 확인된 소목, 오배자 및 황금추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 각각의 추출물을 0, 100, 300 및 500ppm의 농도로 첨가하여 균주의 증식을 조사한 결과 배양후 24시간에 소목 추출물 무첨가구의 OD620값이 0.4인 반면 100ppm 이상의 추출물 첨가 시 0.02∼0.03정도로 균증식이 현저히 억제되었고, 오배자 추출물 무첨가구의 OD620값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 0.02∼0.03정도로 균증식이 현저히 억제되었으며 황금 추출물 무첨가구의 OD620값이 0.4인 반면 300ppm 이상의 추출물 첨가 시 0.02∼0.07 정도로 균증식이 현저히 억제되었다. 이상의 결과로 볼 때 제대염, 부종성 피부염 및 관절염 등의 다양한 가축질병을 일으키는 Staphylococcus aureusi의 감염을 예방하거나 또는 치료할 수 있는 한약재 추출물 사료첨가제의 개발 가능성을 확인 할 수 있었다.

영문 초록

Antimicrobial activity of 18 different traditional medicinal herbs extracts against Staphylococcus aureus was determined by a paper disc method. The Prunella vulgaris, Caesalpinia sappan and Rhus javanica extracts in 5 mg/ml, Poncirus trifoliata, Lonicera japonica and Seutellaria baicalensis extracts in 10 mg/ml and Schizandra chinensis, Alpinia katsumadai, Siegesbeckia orientalis extracts in 30 mg/ml showed a significant antimicrobial activity against Staphylococcus aureus. Minimum inhibitory concentrations of medicinal herbs extracts were in the range of 1∼34mg/ml and 1∼46mg/ml, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. Optical density at 620nm after 24 hours incubation of Staphylococcus aureus in the presence of 100, 300 or 500 ppm of Caesalpinia sappan extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Caesalpinia sappan extract, indicating that growth of Staphylococcus aureus was significantly inhibited within 24 hours by the addition of at least 100 ppm of Caesalpinia sappan extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Rhus javanica extract ranged from 0.02 to 0.03 compared to 0.4 in the absence of Rhus javanica extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Rhus javanica extract. Optical density at 620 nm after 24 hours incubation of Staphylococcus aureus in the presence of 300 ppm of Seutellaria baicalensis extract ranged from 0.02 to 0.07 compared to 0.4 in the absence of Seutellaria baicalensis extract, indicating that growth of Staphylococcus aureus was also significantly inhibited within 24 hours by the addition of at least 300 ppm of Seutellaria baicalensis extract. In conclusion, these findings suggest that extracts from medicinal herbs may play important roles for antimicrobial activities against Staphylococcus aureus.
