Expressional Changes of c-fos, c-jun, and c-myc Induced by 12-O-tetradecanoylphorbol 13-acetate (TPA) in HaCaT Cells

국문 초록

세포의 성장, 분화 및 변형에 관여하는 신호전달체계 중 proto-oncogenes (c-fos, c-jun and c-myc)은 핵 내의 주요한 표적이다. 저자는 정상적인 사람각질화세포주인 HaCaT 세포들에 12-O-tetradecanoylphorbol 13-acetate (TPA)를 2시간, 6시간 처리하여 이 약제에 의한 proto-oncogene들의 발현변화를 조사하였다. Proto-oncogenes들의 변화는 단백질 수준에서는 면역형광염색으로, mRNA는 northern blot으로 확인하였다. 세포주기에 따른 proto-oncogene들의 분포는 유세포분석으로 검증하였다. HaCaT 세포에서 TPA는 단백질과 mRNA 수준 모두에서 c-fos와 c-jun의 발현을 증가시켰고, c-myc의 발현은 감소시켰다. S-G2-M기에서 c-Fos의 경우 2시간 이내에 나타났지만, c-Jun은 점차 유발되었다. G0-G1기에서 c-Myc은 억제되었다. 이 연구결과에서 볼 때 TPA에 노출된 HaCaT 세포들에서 생긴 proto-oncogene들의 발현변화는 이 세포의 성장, 분화 및 변형에 영향을 미칠 것으로 생각된다.

영문 초록

Proto-oncogenes (c-fos, c-jun, c-myc) are major nuclear target for signal transduction pathways involved in the regulation of cell growth, differentiation, and transformation. To investigate expressional changes of proto-oncogene induced by 12-O-tetradecanoylphorbol 13-acetate (TPA), the author treated normal human keratinocyte cell line (HaCaT cells) with TPA for 2 hours and 6 hours. To observe changes of the proto-oncogenes at protein and mRNA levels, immunoflourescence staining and northern blot were used, respectively. To assess the distribution of them during the cell cycle, flow cytometric analyses were performed. TPA increased the expression of c-fos and c-jun, while it decreased the expression of c-myc both at protein and mRNA levels in HaCaT cells. The induction of c-Fos in S-G2-M phase appeared within 2 hours. c-Jun was gradually induced during S-G2-M phase. The inhibition of c-Myc was found in G0-G1 phase. These data suggest that aberrations in the proto-oncogenes expression of HaCaT cells exposed to TPA may contribute to the growth, differentiation, and transformation.
